چکیده

Enteric neurons are able to alter their neurotransmitter content during adaptation to new artificial conditions. The aim of the present study was to investigate changes in vasoactive intestinal peptide (VIP), substance P (SP) and galanin expression during culture of myenteric neurons from the ovine abomasum. In order to accurately reflect the in vivo situation cryostat sections from the ovine abomasum were used. Cultured and non-cultured myenteric neurons were immunohistochemically stained with a mixture of antibodies raised against Hu C/D (neuronal marker) and VIP, SP or galanin. Double labeling revealed that Hu C/D-IR/VIP-IR myenteric neurons were very rare in cryostat sections (1.4 ± 0.2%) but significantly increased to 21.3 ± 1.7% when cultured for three days. A significant increase in Hu C/D-positive/VIP-positive myenteric neurons were also found in 6and 9-days cultures (23.9 ± 1.9 and 24.5 ± 2.0%, respectively). In vivo, the expression of SP was found in 9.7 ± 1.0% of myenteric perikarya. After 3, 6 and 9 days of incubation the proportion of Hu C/D-IR/SP-IR myenteric perikarya significantly increased to 19.3 ± 1.3%, 22.3 ± 1.2% and 24.1 ± 1.7% (respectively). When compared to the in vivo situation the proportion of galanin-expressing myenteric neurons was unchanged after 3, 6 and 9 days of culturing. In conclusion, alterations in VIP and SP (but not galanin) expression occur during neuronal culturing. Our results supports the idea that both VIP and SP may act as factors which increase neuronal survival.

Expression of corticotropin releasing factor (CRF) in the enteric nervous system of the jejunum of sheep

Corticotropin releasing factor (CRF), a 41-amino acid neuropeptide widely distributed in the mammalian central nervous system, has been shown to infl uence several gastrointestinal functions. Recent studies show that CRF released locally from enteric nerves may also underlie alterations in gut function. In this study, immunohistochemisty was applied to demonstrate the presence of CRF in the jej...

Changes in the expression of tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), galanin (GAL), vasoactive intestinal polypeptide (VIP) and substance P (SP) in the uterine cervix-projecting neurons located in the lumbar paravertebral ganglia of the pig.

The uterine cervix-projecting neurons located in the lumbar paravertebral ganglia were identified by retrograde tracing. These contained immunoreactivity to TH and DBH. No immunoreactivity to GAL, VIP and SP was found in the neurons. Extirpation of the uterus reduced the expression of TH and induced the expression of GAL in the neurons. Expression of other substances studied was unchanged.

Myenteric plexus of obese diabetic mice (an animal model of human type 2 diabetes).

The myenteric plexus of the gastrointestinal tract was investigated in the obese diabetic mouse, an animal model of human type 2 diabetes. Sections were immunostained by the avidin-biotin complex method, using a general nerve marker, protein gene product 9.5 (PGP 9.5), as well as antibodies to several important neurotransmitters. Computerized image analysis was used for quantification. When dia...

The distribution, colocalisation, and interconnections of nitrinergic and peptidergic neurons and nerves in the human oesophagus were examined. Cryosections of surgically resected tissues from eight subjects were studied with indirect immunofluorescence for the presence of 11 neuropeptides and neuron specific enolase. After immunohistochemistry, nitric oxide synthase was shown on the same secti...
